General description

Interleukin-6 (IL-6) mediates its biological effects through the type I IL-6 receptor system that consists of two chains, IL-6Rα and gp130. The IL-6Rα chain is the binding component specific to IL-6; while the gp130 only transmits signals of IL-6 when bound to IL-6Rα. IL-6R has a role in various signaling cascades. The gp130 also can transmit signals from leukemia inhibitory factor (LIF), oncostatin M (OSM), ciliary neurotrophic factor (CNTF), interleukin-11 (IL-11) and cardiotrophin-1 (CT-1) in conjunction with other receptor subunits. The low-affinity binding site for IL-6 is composed of IL-6Rα alone. IL-6Rα is expressed in a wide range of cells including T cells, fibroblasts and macrophages. Soluble IL-6Rα which consists of only the extracellular domain of the IL-6Rα chain, acts as an agonist of IL-6 activity at low concentrations. Recombinant human sIL-6Rα is a 37.6kDa protein consisting of the extracellular domain of the IL-6Rα chain (339 amino acid residues).

Application

sIL-6 Receptor alpha human has been used in cell culture to treat fibroblasts. It has also been used in stimulation experiments to analyze IL-6 trans-signaling.

Biochem/physiol Actions

Interleukin-6 Receptor Alpha (IL-6Rα) plays a dual role in signaling. In classical signaling, it interacts with glycoprotein 130 to promote the anti-inflammatory and regenerative effects of IL-6. In contrast, during trans-signaling, it facilitates the pro-inflammatory effects of IL-6 by allowing cells lacking IL-6Rα to respond to IL-6 in the presence of soluble IL-6Rα. IL-6Rα exists in both soluble and transmembrane forms. The transmembrane form is integrated into the cell membrane of target cells. IL-6 and the IL-6 receptor alpha chain (IL-6R) are expressed by various cell types, including monocytes, lymphocytes, fibroblasts, and endothelial cells.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/ml. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20°C to -80°C.
