General description

The protein named Transcription factor EB (TFEB) or alternatively in older literature, Class E basic helix-loop-helix protein 35 (bHLHe35) and encoded by the human gene TFEB/BHLH35 is a transcription factor that specifically recognizes and binds E-box sequences (5′-CANNTG-3′) on DNA. Efficient DNA-binding requires dimerization with itself or with another MiT/TFE family member such as TFE3 or MITF. TFEB in association with the protein TFE3, activates the expression of CD40L in T-cells, thereby playing a role in T-cell-dependent antibody responses in activated CD4+ T-cells and thymus-dependent humoral immunity. TFEB also plays a central role in expression of lysosomal genes. TFEB specifically recognizes and binds the CLEAR-box sequence (5′-GTCACGTGAC-3′) present in the regulatory region of many lysosomal genes and through binding acts as a positive regulator of their expression. Finally, TFEB appears to be a positive regulator for autophagy related gene expression, heavy chain immunoglobulin gene expression, and genes necessary for the proper vascularization of important tissues such as the placenta.

Application

Detect TFEB using this rabbit polyclonal antibody, Anti-Transcription factor EB Antibody validated for use in western blotting & IP.
Immunoprecipitation Analysis: 10 µg from a representative lot immunoprecipitated Transcription factor EB in 500 µg of Raji cell lysate. Immunopecipitated Transcription factor EB was then detected in Western Blot.

~58 kDa and ~40 kDa observed. Uniprot describes two isoforms at 44 kDa and 53 kDa This protein has been observed at 44-53 kDa due to sumoylation (Miller, A. J., et al. (2005). J Biol Chem. 280(1):146-55.).
