Mouse Orexin-A. Several peptides associated with feeding behavior have been reported recently. Orexins (Orexin-A and Orexin-B) are a family of hypothalamic neuropeptides selectively expressed in the hypothalamus (Sakurai et al. 1998). Orexin-A and Orexin-B are derived from the same precursor (Prepro-orexin) by proteolytic cleavage. Prepro-orexin is an 130 aa peptide with a putative 33 aa secretory sequence, a hydrophobic core followed by residues with small polar side chains. The expression was detected in brain and to a small extent in testis (Sakurai et al. 1998). These neuropeptides bind and activate two closely related Orexin receptors--G protein coupled receptors (GPCRs) OX1R and OX2R.
Immunogen
An 18 amino acid peptide mapping near the amino terminus of mouse Orexin-A. The immunogen sequence is identical in human, rat, porcine, ovine, bovine and canine and has 81% homology with chicken.

Western blot: 1-10 μg/mL using Chemiluminescence technique ELISA: 1:10,000 - 100,000 using 5-100 ng of control peptide per well. NOTE: Orexin A is a 3.5kDa protein. Adjust your SDS-PAGE gel and lane size markers accordingly.
Immunohistochemistry: Frozen 4% PFA tissue with ABC detection {Wagner et al 2000}.
Optimal working dilutions must be determined by end user.
Physical form
Liquid in PBS, pH 7.4 with 0.1% BSA and 0.1% sodium azide.
Storage and Stability
Maintain frozen at -20°C in undiluted aliquots for up to 12 months. Avoid repeated freeze/thaw cycles.
