ZFN 是一类工程 化的DNA 结合蛋白，可导致双链断裂。这种双链断裂是位点特异性诱变介导的 DNA 修复所必需的。它通过与用户指定的基因座结合来促进靶向基因组的编辑。谷氨酰胺合成酶（GS）是生物制药行业中最常用的选择性标记之一。 谷氨酰胺是细胞生长的必需氨基酸。GS酶负责将谷氨酸盐转化为谷氨酰胺。没有这种酶活性的话，细胞不能再合成内源谷氨酰胺，如果培养基中不补充谷氨酰胺，细胞将死亡。
利用 Sigma ′CompoZr 锌指核酸酶 (ZFN) 专利技术创建 CHOZN 细胞系，在亲代 CHO-K1 细胞中创建遗传修饰。 细胞储存在化学成分明确、无动物成分的 EX-CELL CD CHO 融合培养基中

細胞系來源

CHOZN 细胞系均为 1957 年 Puck 创立的亲本 CHO-K1 细胞系的亚克隆。
