重组酶cre (UniProt:P06956;也称为Cre)由在大肠埃希菌噬菌体P1(噬菌体P1)中的Cre基因编码(基因ID:2777477)。Cre(引起与环化重组酶的重组)是′噬菌体′整合酶家族的38 kDa DNA重组酶,来源于P1噬菌体。它催化位点特异性重组,并对P1 DNA中发现的34 bp DNA序列基序loxP(X-over P1位点)具有高度特异性。loxP位点由两组13 bp回文序列(ATAACTTCGTATA-NNNTANNN-tatacgaagtat)之间的可变不对称8-bp序列组成。两个cre重组酶蛋白,每个结合到loxP位点的两个13 bp回文序列中的一个,形成二聚体。然后,该二聚体与另一个loxP位点上的第二个二聚体结合形成四聚体。由四聚体连接的两个位点在方向上平行,双链DNA在两个loxP位点均被切割。然后用DNA连接酶重新连接这些链。由于Cre基因和loxP位点不是大多数物种所固有的,它们通常被引入靶细胞或组织中,并被用作一种手段来精确控制体外培养细胞和体内动物中的基因表达。
