一般說明

Notch是单程跨膜受体蛋白家族,在内质网中以无活性形式合成,并在识别后被反式高尔基体网络中的弗林蛋白酶样转化酶(S1裂解)在细胞外部位点进行蛋白水解裂解。RQRR序列在以异二聚体的形式到达质膜之前产生活性的,配体可及的形式。Notch家族由4个成员(1-4)组成,其配体包括Delta和Jagged配体家族。这些配体引起Notch的蛋白水解以释放细胞内结构域。切割产生C末端片段N(TN)和N末端片段N(EC)。配体结合后,它被TNF-α转化酶(TACE)裂解(S2裂解),产生与膜相关的中间片段,称为Notch细胞外截短(NEXT)。然后,该片段被gly1743和val1744之间的早老素依赖性γ-分泌酶切割(S3切割),以从膜释放细胞内结构域(NICD)。该结构域易位至细胞核并通过结合DNA结合蛋白Csl启动转录事件。Notch家族成员参与细胞的分化和发育。
