mTOR(雷帕霉素的哺乳动物靶标,又名 FRAP、RAPT 或 RAFT)是 289kDa 的 Ser/Thr 蛋白激酶,可调节细胞周期进程、细胞生长、蛋白合成、核糖体生物合成和自噬。mTOR 是磷酸肌醇激酶相关激酶(PIKK)家族的进化保守成员,其活性通过响应胰岛素或肌肉活性,由 Akt 在 Ser2448 上的磷酸化来调节。 mTORC1(mTOR复合物 1)至少由mTOR、Raptor和G(mLST8)组成。众所周知,mTORC1在胰岛素信号传导中发挥核心作用,而胰岛素信号传导对维持代谢平衡至关重要。mTORC2主要通过使Ser473上的疏水基团Akt(PI3激酶通路的著名效应器)磷酸化来影响细胞的增殖和存活。除了磷酸化 Akt 外,还已知 mTORC2 复合物通过 Rac、Rho 和 PKC 发挥其作用,影响细胞骨架的组织和迁移。两种 mTOR 复合物中的缺失都与多种疾病有关,包括癌症和糖尿病。
