General description

TNFSF12 (tumor necrosis factor, member 12) is a novel member of the tumor necrosis factor (TNF) ligand family. The multifunctional cytokine, TNFSF12 is widely distributed in human tissues.

Application

Biochem/physiol Actions

TNFSF12 (tumor necrosis factor, member 12) plays a key role in the tissue regeneration and remodeling. It binds to the short C-terminal cytoplasmic tail of the cognate receptor Fn14 to accelerate signaling phenomena. TNFSF12-Fn14 interaction increases the therapeutic value of both the ligand and receptor. It also acts as an apoptotic inducer in association with interferon-γ or cycloheximide. High impact of TNFSF12 on the myocardial functions has been reported. Alteration in the TNFSF12 gene leads to the metabolic syndrome (MetS) and high cardiovascular risk. It has been suggested that TNFSF12 may play a vital role in the formation of vasculature in vivo.
