General description

Tetramethylrhodamine isothiocyanate (TRITC) is stable and less sensitive to light compared to fluorescein isothiocyanate (FITC). The conjugate of albumin with TRITC is effective for following microcirculation in vivo microscopy. Albumin-TRITC aids tracking of calcium phosphate nanoparticle bound DNA in human umbilical vein endothelial cells (T-HUVEC).

Application

Albumin, Tetramethylrhodamine isothiocyanate has been used:
  • in photoinduced release assay of polyethylene beads
  • for passivating poly(methyl methacrylate) (PMMA) layer prior to immunoglobulin G coating
  • in fluorescent correlation spectroscopy

Red-fluorescing TRITC-albumin was used to track the uptake of calcium phosphate/DNA nanoparticles during cell transfection. The dye-linked albumin is incorporated into the nanoparticles. Efficiency of transfection could be followed by fluorescence microscopy.

Preparation Note

Prepared from crystallized and lyophilized albumin (A4378).
