Avoid cell culture media containing NO3, such as Fe(NO)3 present in DMEM/F12. Centrifuge cell or tissue culture media at 1000 * g for 15 minutes to remove insoluble material. Collect the supernatant for testing. Serum or plasma should be centrifuged in an Amicon® Ultra-4 Centrifugal Filter Unit with Ultracel®-10

The Griess assay is one of the most popular and simplest methods used to detect the NO concentration. The Griess assay′s mechanism is summarized as the azo coupling between diazonium species, which are produced from sulfanilamide with NO2, and naphthylethylenediamine. The NO2/NO3 Assay Kit contains these dyes, a nitrate reductase, an enzyme co-factor, buffer solution and NO2, NO3 solutions as standards. Therefore, total NO metabolites are easily detectable using this kit. The suitable NO2 detection range is from 10 to 100 μM.

How can I determine the shelf life / expiration / retest date of this product?

If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

How is shipping temperature determined? And how is it related to the product storage temperature?

Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers. Can I use a frozen cell culture supernatant or must it be obtained on the same day for the assay?
1 answer-
When using frozen samples, it is recommended to thaw the samples completely, mix well by vortexing and centrifuge prior to use to remove particulates. Note that cell culture medium that contains NO3- as a component (such as RPMI-1640) are not a suitable to detect [NO3- + NO2-]. However, NO2- in the culture medium can be determined using this kit.
