General description

Tankyrase-1/TNKS is a critical enzyme in Wnt signaling pathway. Tankyrase-1/TNKS is a poly-ADP-ribosyltransferase which is a key mediator of PARsylation: Becoming ADP-ribosylated. PARsylation is important in Wnt signaling because ADP-ribosylation of AXIN1 and AXIN2, two key components of the beta catenin destruction complex, is necessary for destruction via RNF146 (an ubiquitin lyase) and ubiquitination and subsequent destruction of beta catenin. Tankyrase-1/TNKS also mediates PARsylation of TERF1 and thus is important in controlling telomere length, and Tankyrase-1/TNKS’s PARsylation of other proteins makes Tankyrase-1/TNKS important in vesicle trafficking and subcellular protein distribution as well. Tankyrase-1/TNKS is widely expressed, and is found mostly cytoplasmically but can be associated with the nucleus and spindles and Golgi complexes as well.

Application

Immunocytochemistry Analysis: A 1:50 dilution from a representative lot detected Tankyrase-1/TNKS in Hela, A431, and N1H/3T3 cells.

Immunohistochemistry Analysis: A 1:250 dilution from a representative lot detected Tankyrase-1/TNKS in neurons and glial cells in human cerebral cortex tissue, Purkinje, basket and granule cells in human cerebellum tissue, and neurons in human thalamus tissue.

This Anti-Tankyrase-1/TNKS Antibody, clone 7D1.2 is validated for use in Western Blotting and Immunocytochemistry and Immunohistochemistry for the detection of Tankyrase-1/TNKS.

Quality

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: 4 µg/mL of this antibody detected Tankyrase-1/TNKS in 10 µg of HeLa cell lysate.

Target description

~142 kDa observed.
Uncharacterized bands may be observed in some lysates.
