General description

Vimentin belongs to the class of type III intermediate filament protein family. The gene is located on human chromosome 10p13. It is a cytoskeletal protein. Vimentin is mainly expressed in normal mesenchymal cells.
Vimentin is a 58 kD class III intermediate filament present mainly in mesenchymal cells. It regulates cell shape, motility and endurance of mechanical stress in mesenchymal cells. Monoclonal anti-vimentin antibody can be used for the localization of vimentin by various immunochemical assays like immunoblotting, immunohistochemistry and immunoprecipitation. Frozen B5 fixed and formalin fixed, paraffin embedded tissue sections can be stained using anti-vimentin antibody. Mouse monoclonal anti-vimentin antibody reacts specifically with an epitope of the cytoskeletal protein vimentin (58kD). The product shows cross reactivity with vimentin from bovine, human, pig, sheep, cat, rat, mouse and rabbit.

Specificity

Recognizes an epitope of the cytoskeleton protein vimentin that is not detected in human lymphoid cells or derived malignancies. It is therefore useful in differentiating sarcomas from lymphomas.

Application

Monoclonal Anti-Vimentin antibody can be used in immunocytochemistry and immunohistochemical staining. It can also be used in immunofluorescence staining (diluted 1: 500) .
Monoclonal Anti-Vimentin antibody produced in mouse has been used in western blotting and immunolabeling.

Biochem/physiol Actions

Vimentin plays an important role in modulating cell growth. It plays a pivotal role in epithelial-mesenchymal transition. Vimentin acts as a potential target for anti-metastasis therapy for nasopharyngeal carcinoma (NPC). It regulates cellular architecture and tissue integrity. It is also involved in tumorigenesis and the metastatic spread of cancer. Vimentin provides stress resistance. It might be involved in neuritogenesis.
