General description

Streptavidin is a biotin-binding protein.[1] Horseradish peroxidase (HRP) is a heme-containing enzyme.[2] HRP-Streptavidin consists of the protein streptavidin, as a secondary detection reagent, conjugated to the enzyme HRP.[3] The conjugate is used in ELISA to increase the detection of various kinds of targets.[4]

Application

HRP-Streptavidin has been conjugated to secondary antibodies for western blotting.[5]

Questions

1–3 of 3 Questions
  1. What is the product concentration for RABHRP3?

    1 answer
    1. The product concentration for RABHRP3 is 600 µL, which is a 100X concentrated HRP-conjugated streptavidin. It should be diluted 1:100 in an appropriate diluent before use.

      Helpful?

  2. I have a question about RABHRP3. I need to know the appropriate dilution to use for this product.

    1 answer
    1. The RABHRP3 Streptavidin-HRP Conjugate is specifically intended for titered use in RAB series ELISA only. The appropriate dilution factor for the kit it is used in will be listed in the ELISA protocol.

The exact concentration is not known and may vary from lot to lot to ensure performance in the ELISA kit. If this product is not being used in an RAB series ELISA, it would be advisable to use a different HRP conjugate.

We have several alternative options available, but cat# 18-152 is commonly used for ELISA and Western Blots and is provided at a 1 mg size.

      Helpful?

  3. What is the difference between HRP-Strepatavidin, HRP conjugate? My application includes use in ELISA method.

    1 answer
    1. The RABHRP3 HRP-Streptavidin product is a conjugate of the two components. It is optimized for use in ELISA kits with product numbers beginning with the letters RAB. The following link displays a list of these kits: https://www.sigmaaldrich.com/US/en/search/rab*-elisa?focus=products&page=1&perpage=30&sort=relevance&term=rab%2A%20elisa&type=product

Other HRP-Peroxidase conjugate products have not been optimized specifically for use with these kits. In addition to HRP-Peroxidase conjugate products, other pairings are available. These include secondary antibodies, Protein A & G, and lectins. For ELISA applications, secondary antibodies are the most prevalent.
