General description

Biochem/physiol Actions

Tumor necrosis factor receptor 2 (TNFR2) plays a vital role in inducing signals for various cellular process, including proliferation of thymocytes and peripheral T cells, T cell secretion of granulocyte-macrophage colony-stimulating factor, inhibition of early hematopoiesis, and the inactivation and clearance of TNF by the kidney. Mutation in the gene leads to hematopoietic cell malignancies, neuroblastoma, glioma, cervical and ovarian carcinoma. TRAF2 interacts with TNFR2 and activates NF-kappa B (NF-κB) pathway. TNFR2, which lacks death domains, modulates the effect of other death receptor TNFR1, which induces T cell death. TNFR2 plays an essential role in TNF-α–induced leukocyte-endothelial interaction. TNFR2 is considered as a potential therapeutic alternative, as it has capacity to nullify the common side effects associated with drugs targeting TNF-α or p55.
