General description

Casein is a milk protein, that is not expensive and nontoxic. highly stable.[1] This phosphoproteins accounts for 80% of the total protein present in bovine milk. Casein is a member of one of the larger family of secretory calcium?binding phosphoproteins.[2]

Application

Casein, N,N-dimethylated from bovine milk has been used:
  • in the determination of general proteolytic activity[3]
  • as a substrate to study the effect of dephosphorylation on the calcium-activated neutral proteases (CANP) proteolysis of paired helical filaments (PHF) II-τ[4]
  • in transglutaminase (TG) assay[5]

Casein, N,N-dimethylated from bovine milk can be used in assays to detect and help determine the functional role of tissue transglutaminase (tTG) in disease. In particular, Casein, N,N-dimethylated has been used in the mechanic and kinetic analysis of tTG on protein substrates, which may be linked to neurodegenerative disease.

This preparation has less than 10% reactivity with 2,4,6-trinitrobenzenesulfonic acid (TNBS) compared to non-methylated casein.

Protease substrate that has been used to assay basilase[6] and a subtilisin analog that had been produced by site-directed mutagenesis.[7]

Biochem/physiol Actions

Casein is highly preferred as a drug delivery vehicle.[1] It possess antioxidant activity.[8]

    1. The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.

    1. This product is not sold by purity, but by the percent of N,N-methylation. It is required to be at least 90% methylated.

    1. Unfortunately, we have not determined the molecular weight. The molecular weight of casein is 26 kDa or less; the molecular weight of Product No. C9801 is expected to be a bit higher.

    1. The solubility has been tested at 10mg/mL in 0.1M Boric acid, resulting in a white haze.
