General description

Tn antigen is a truncated O-glycan, that is commonly noticed in colorectal cancer (CRC). It is usually expressed at high levels in cancer cells. Tn antigen is small in size and simple in structure. It has a non-physiological glycan structure in humans, hence it may be identified as foreign by the immune system.

Biochem/physiol Actions

Tn antigen stimulates colorectal cancer metastasis through the activation of epithelial-mesenchymal transition, mediated by H-Ras. It acts as a precursor to several significant complex O-glycans. Subpopulations of blood cells in all lineages carrying the Tn antigen results in a hematological disorder called Tn syndrome.

The study describes development of a glycan biosensor for detection of a tumor-associated antibody. The glycan biosensor is built on an electrochemically activated/oxidized graphene screen-printed electrode (GSPE). Aberrant O-glycosylation truncates O-glycans and is known to be closely associated with colorectal cancer (CRC), a major gastrointestinal tumor. Expression of the tumor-associated glycan Tn antigen (αGalNAc-Ser/Thr) has been correlated to poor prognosis and metastasis in multiple cancer types. However, the exact mechanisms exerted by Tn antigen to support tumor growth are still lacking. One emerging hallmark of cancer
