General description

Embryonic Stem Cell line derived from C57BL/6N blastocysts. C57BL/6N is the most commonly used inbred mouse strain. Large scale mouse knockout projects such as NIH’s KOMP project use C57BL/6 as the standard background for mouse genome engineering. C57BL/6 is also the reference strain for mouse genome sequencing at the NHGRI. C57BL/6N mice show good breeding performance. As a commonly used reference strain, transgenic mouse models are often backcrossed to a uniform C57BL/6 background, and B6 ES cells facilitate building complex models which combine gene inactivation with existing transgenics maintained in a B6 background. Additional phenotypic information is available at the Mouse Genome Informatics website (http://www.informatics.jax.org/external/festing/mouse/docs/C57BL.shtml). The B6 ES line was genome verified by determining the alleles at 29 SNPs that are informative across a large number of inbred mouse strains.

Spermatogonial stem cells (SSCs) originate from gonocytes that differentiate from primordial germ cells (PGCs). In the developing mouse testis, expression of the gene LIM homeobox 1 (Lhx1) marks the most undifferentiated SSCs, which has not yet been reported for spermatogonia-like
