Application

p-Nitrophenyl phosphate (p-NPP) is the customary product utilized for the assay of alkaline phosphatase. It is usually supplied in tablet or powder form and is dissolved in an appropriate buffer immediately prior to assay. Liquid p-NPP concentrates are also available, but must also be diluted in buffer. CHEMICON, Inc. now provides p-NPP stabilized in diethanolamine (DEA) buffer and available for instant use. The DEA buffer contains Mg++ and Zn++ ions providing optimum activation of alkaline phosphatase labeled probes.

This is an excellent substrate for phosphatase based ELISA assays.

It produces a soluble end product which is yellow in color and reads at 405 nm to 410 nm. Both kinetic and end point determinations can be performed.
