Description générale

Le nouveau kit de dosage des triglycérides (réf. MAK564) est désormais disponible ! Les triglycérides (TG) sont les principaux constituants des huiles végétales, des graisses animales, des LDL et VLDL, et jouent un rôle important comme transporteurs d′acides gras ainsi que source d′énergie.[1][2] Les TG sont décomposés en acides gras et en glycérol, après quoi ces deux constituants peuvent servir de substrat pour les voies métaboliques et productrices d′énergie.[3] L'hypertriglycéridémie est associée à l'athérosclérose, aux cardiopathies et aux accidents vasculaires cérébraux ainsi qu'à la pancréatite.[4]

Adéquation

Ce produit peut être utilisé avec les échantillons de sérum, plasma et autres liquides biologiques, ainsi qu′avec les échantillons de culture cellulaire et tissulaire.

Principe

Le kit de quantification des triglycérides propose une méthode simple et sensible pour mesurer la concentration en TG de divers types d′échantillons. Dans cet essai, les TG sont convertis en acides gras libres et en glycérol. Le glycérol est alors oxydé en un produit mesurable par colorimétrie (570 nm) / fluorimétrie (lex = 535 nm/ lem = 587 nm). Ce kit permet de détecter entre 2 pmoles et 10 nmoles (plage de 2 à 10 000 mM) de triglycérides dans divers types d′échantillons. Ce kit détecte également les monoglycérides et les diglycérides.

  1. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  3. Can ethanol or isopropanol (IPA) be used instead of water for liver tissue sample preparation with the Triglyceride Quantification Colorimetric/Fluorometric Kit (MAK266)?

    1. Based on the guidelines, only water is an appropriate solvent for samples in this assay. Ethanol and isopropanol are not suitable solvents for this assay.

  4. Is liver tissue extracted using RIPA buffer and without NP-40 compatible with the MAK266 Triglyceride Quantification Colorimetric/Fluorometric Kit?

    1. It is recommended to use NP-40 or similar non-ionic detergents instead of RIPA, due to the presence of harsh detergents in RIPA.

    Terrible kit!

    The protocol is not well-written. I looked for several literatures who used this kit and found inconsistent data for control samples. Even the technical staffs of Sigma could not satisfy my questions and I strongly discourage other shoppers to use this.
