General description

Type 2 protein serine/threonine phosphatase (PP2A) is a trimer composed of a catalytic C subunit and two regulatory (A and B) subunits; many isoforms for the A and B subunits have been found. PP2A has a high specific activity against the α subunit of phosphorylase kinase and is insensitive to Inhibitor-2. PP2A is distinguishable from PP2B/Calcineurin and PP2C by its partial activity in the absence of divalent cations, and by its high sensitivity to inhibition by okadaic acid. It is also thought to be the principal downmodulator of serine/threonine kinase cascades. PP2B requires Ca2+/calmodulin for activity, and is an activator of NFAT transcription factors. PP2C requires Mg2+, but is insensitive to okadaic acid, microcystin LR, or the calmodulin inhibitor trifluoperazine.

Specificity

This antibody recognizes PP2A catalytic subunit, Mr 36 kDa.

Immunogen

A 16 residue synthetic peptide (C-RGEPHVTRRTPDYFL) corresponding to amino acids 295-309 of the 36 kDa catalytic subunit of human protein phosphatase 2A (PP2A). Clone 1D6.
Epitope: a.a. 295-309

Application

Detect PP2A using this Anti-PP2A Antibody, C subunit, clone 1D6 validated for use in IC, IP & WB.
Immunoprecipitation:
4 µg of a previous lot immunoprecipitated PP2A from 500 µg of A431 RIPA lysate. Note: This antibody has been reported not to block phosphatase activity.

Immunocytochemistry:
5-10 µg/mL of a previous lot of this antibody showed positive immunostaining for PP2A in A431 cells fixed for 10 minutes with 4% paraformaldehyde in PBS.

Quality

Routinely evaluated by western blot on human A431 or murine 3T3/NIH RIPA cell lysates.

Western Blot Analysis:
0.5-2 µg/mL of this lot detected PP2A in human A431 and previously in murine 3T3 RIPA cell lysates.

Target description

36 kDa

Physical form

Format: Purified
Protein A chromatography
Purified mouse monoclonal IgG2bκ in buffer containing 0.1M Tris-Glycine, pH 7.4, 0.15 M, NaCl, and 0.05% sodium azide. Frozen solution.

Storage and Stability

Stable for 1 year at -20ºC from date of receipt. Aliquot to avoid repeated freezing and thawing. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Analysis Note

Control
Positive Antigen Control: Catalog #12-301, non-stimulated A431 cell lysate. Add 2.5µL of 2-mercaptoethanol/100µL of lysate and boil for 5 minutes to reduce the preparation. Load 20µg of reduced lysate per lane for minigels.

Other Notes

Concentration: Please refer to the Certificate of Analysis for the lot-specific concentration.
