Description générale

Standardization of sample analysis is currently needed in microbiome genomics research workflow. Lack of standardization can lead to biases and errors in common processes during sample preparation and analysis such as sample amplification, sequencing and bioinformatics analyses.1 Thermobifida fusca genomic DNA microbial standard can serve as standard for benchmarking the performance along the workflow of microbiomics or meta-genomics analyses and as a tool to increase reproducibility and allow comparison of results obtained by different labs.T. fusca, is a moderately thermophilic gram-positive soil bacterium, that belongs to actinobacteria, with optimum growth at 55-60°C.2,3 The genome of T. fusca has a high GC content (67.5%) 3 and therefore it may serve as a quality marker for the PCR stage in the NGS workflow. It is a major degrader of plant cell walls and has been used as a model organism for the study of secreted, thermostable cellulases. 3 Thermostable enzymes from T.fusca have been reported to replace thermochemical pre- treatments of cellulosic biomass in the process of decomposition to biofuels.4T. fusca, being a soil bacterium, is not likely to be found in the human microbiome. Therefore, T. fusca genomic DNA standard can be used as an internal standard for the NGS workflow; as a negative control for sample preparation, or as a reference DNA for sequencing.
