General description
Mycoplasma Broth is a basal medium which, after enrichment with yeast extract, horse serum, and antibiotics, will support the growth of Mycoplasma sp. The components contained in this basal medium do not exhibit inhibitory or toxic effects when used to culture mycoplasma. A special mineral supplement has been added to improve the growth and colony characteristics of the organism.
Application
Mycoplasma Broth has been used for isolation and cultivation of mycoplasma. It may also be used to grow Porphyromonas gingivalis under anerobic conditions.
Preparation Note
Basal Medium:
1. Suspend 25.5 g of powdered medium in 1L of distilled water. Stir until the powder dissolves.
2. Adjust the pH of the solution to 7.8 ± 0.2.
3. Autoclave at 121 °C and 15 psi for 15 minutes to sterilize. Cool to 50 °C and add sterile supplements.

Store dry powdered medium at room temperature and prepare medium at 2-8 °C.
Other Notes
Powder contains (g/L): 10.0 bacterial peptone, 10.0 LAB-LEMCO powder, 5.0 sodium chloride, 0.5 mineral supplement.
