General description

UDP-glucuronosyltransferase 1-10 (UniProt: Q9HAW8; also known as EC: 2.4.1.17, UDPGT 1-10, UGT1*10, UGT1-10, UGT1.10, UDP-glucuronosyltransferase 1-J, UGT-1J, UGT1J, UDP-glucuronosyltransferase 1A10) is encoded by the UGT1A10 (also known as GNT1, UGT1) gene (Gene ID: 54575) in human. UDP-Glucuronosyltransferases (UGTs) are a major group of phase II drug metabolizing enzymes and each enzyme in this family displays a unique substrate specificity and tissue-specific expression patterns. UDP-Glucuronosyltransferases mediate the transfer of glucuronic acid from UDP-glucuronic acid to various aglycone substrates making them more hydrophilic and readily excretable. UGT1A10 is a single-pass membrane protein that is synthesized with a signal peptide (aa 1029), which is subsequently cleaved off in the mature form. Two isoforms of UGT1A10 have been described that are produced by alternative splicing. Both isoforms 1 and 2 are expressed in colon, esophagus, and small intestine. However, in liver only isoform 2 is expressed. Isoform 2 lacks transferase activity, but it can act as a negative regulator of isoform 1. Expression of UGT1A10 is regulated both epigenetically and transcriptionally in response to oxidative stress and aryl hydrocarbon receptor agonists. Clone 20H1 is highly specific for UGT1A10 that can also detect differentially glycosylated forms of UGT1A10. (Ref.: Oda, S., et al. (2017). Drug Metab. Dispos. 45:1027-1034).

Specificity

Clone 20H1 specifically detects human UDP-glucuronosyltransferase 1-10. It targets an epitope with in 32 amino acids from the N-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 32 amino acids from the N-terminal region of human UDP-glucuronosyltransferase 1-10.
