Specificity
Clone 1L16 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Paired mesoderm homeobox protein 1 (PRRX1). It targets an epitope within 21 amino acids from the N-terminal half. Predicted to react with both isoforms.

KLH-conjugated linear peptide corresponding to 21 amino acids from the N-terminal half of human Paired mesoderm homeobox protein 1 (PRRX1).

Target description
Paired mesoderm homeobox protein 1 (UniProt: P54821; also known as Homeobox protein PHOX1, Paired-related homeobox protein 1, PRX-1) is encoded by the PRRX1 (also known as PMX1) gene (Gene ID: 5396) in human. PRRX1 is considered as a master transcription factor in mesenchymal cells in embryos and in certain cancer types. It can serve as a transcription co-activator, enhancing the DNA-binding activity of serum response factor that is required for the induction of genes by growth and differentiation factors. PRRX1 acts as a transcriptional regulator of muscle creatine kinase and has a role in the establishment of diverse mesodermal muscle types. It binds to an A/T-rich element in the muscle creatine enhancer. PRRX1 is reported to orchestrates the functional drift of fibroblasts into myofibroblastic phenotype via TGF-β signaling. These reprogrammed fibroblasts have myofibroblastic functions that result in enhanced tumorigenicity and aggressiveness of cancer. PRRX1 expression in cancer-associated fibroblast (CAF) is reported to have an unfavorable prognosis in multiple cancer types. Fibroblast-specific PRRX1 depletion can induce long-term and sustained complete remission of chemotherapy-resistant cancer in murine models. PRRX1 expression is also observed in quiescent human oligodendrocyte progenitor cells (OPCs) where it induces cell-cycle arrest and inhibits migration. Enforced PRRX1 expression can result in reversible G1/0 arrest. Two isoforms of PRRX1 have been described that are produced by alternative splicing. Although both isoforms are known to reduce OPCs proliferation, only PRRX1a reduces migration. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
