Specificity

Clone 2C17 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects -Actinin-1. It targets an epitope within 22 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the C-terminal half of human -Actinin-1.

Application

Quality Control Testing

Evaluated by Western Blotting in A431 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected -Actinin-1 in A431 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected -Actinin-1 in C6, Cos7, NIH3T3, and Hek293 cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected -Actinin-1 in human small intestine tissue sections.

Immunofluorescence Analysis: A 1:100 dilution from a representative lot detected -Actinin-1 in human small intestine tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Alpha-actinin-1 (UniProt: P12814; also known as Alpha-actinin cytoskeletal isoform, F-actin cross-linking protein, Non-muscle alpha-actinin-1) is encoded by the ACTN1 gene (Gene ID: 87) in human. α-Actinin-1 is a homodimeric, non-muscle cytoskeletal protein that serves as F-actin cross-linker protein and anchors actin to a variety of intracellular structures. It is found along microfilament bundles and adherens-type junctions, where it is involved in binding actin to the membrane. Its actin binding region is localized in amino acids 1-247.  It contains four spectrin repeats and has two calponin-homology (CH) domains (aa 31-135 and 144-250). α-Actinin-1 is reported to promote activity of the L-type Ca2+ channel Cav1.2. It recruits CaV1.2 to defined surface regions and boost its open probability. Overexpression of α-actinin-1 is frequently observed in human breast cancers. In mammary epithelial cells, higher levels of α-actinin-1 promote cell migration  accompanied by a major reorganization of the actin cytoskeleton and the associated E-cadherin-based adhesions.  Mutations in ACTN1 gene are associated with platelet type bleeding disorder (BDPLT15) that causes a reduction in the number of large platelets and mild bleeding tendency. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, Affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
