Specificity

Clone 5K19 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Vascular cell adhesion protein 1 (VCAM-1). It targets an epitope within 17 amino acids from the C-terminal, extracellular domain.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal, extracellular domain of human Vascular cell adhesion protein 1 (VCAM-1).

Application

Quality Control Testing

Evaluated by Western Blotting with human recombinant VCAM-1.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected human recombinant VCAM-1.

Tested applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected VCAM-1 in C2C12, NIH3T3, HUT-78, and MCF-7 cell lysates.

Affinity Binding Assay: A representative lot of this antibody bound VCAM-1 peptide with a KD of 5.6 x 10-6 in an affinity binding assay.

Immunofluorescence Analysis: A 1:1,000 dilution from a representative lot detected VCAM-1 in human tonsil tissue sections.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected VCAM-1 in human tonsil tissue sections.

Anti-VCAM-1, clone 5K19 ZooMAb®, Cat. No. ZRB1132, is a recombinant Mouse monoclonal antibody that specifically targets VCAM-1 and is tested for use in Affinity Binding Assay, Immunofluorescence, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Vascular cell adhesion protein 1 (UniProt: P19320; also known as V-CAM 1, VCAM-1, INCAM-100, CD106) is encoded by the VCAM1 gene (Gene ID: 7412) in human. VCAM-1 is a single-pass type I membrane glycoprotein that is predominantly expressed in endothelial cells. It is reported to be absent on resting endothelial cells, but the cells respond to IL-1 and TNFa by upregulating the expression of VCAM-1, with maximal activity reaching at 6 to 12 hours. VCAM-1 functions in leukocyte-endothelial cell adhesion. It interacts with integrin a4/b1 on leukocytes and mediates both adhesion and signal transduction. Under high degree of inflammation and chronic conditions in some diseases, VCAM-1 expression is also expressed on the surface of other cells, including tissue macrophages, dendritic cells, bone marrow fibroblasts, myoblasts, oocytes, Kupffer cells, Sertoli cells, and cancer cells. VCAM-1 is synthesized with a signal peptide (aa 1-24), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 25-698), a transmembrane domain (aa 699-720), and a cytoplasmic domain (aa 721-739). The extracellular domain contains seven Ig-like C2-type domains. Ig-like domain pairs of 1 and 4, 2 and 5, and 3 and 6 are highly homologous with each other. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
