Specificity

Clone 13F10 is a ZooMAb rabbit recombinant monoclonal antibody that detects p44 MAPK (ERK1). May also display reactivity with p42 MAPK (ERK2).

Immunogen

KLH-conjugated linear peptide corresponding to 35 amino acids from the C-terminal region of rat p44 MAPK (ERK1).

Application

Anti-p44 MAPK (ERK1), clone 13F10 ZooMAb, Cat. No. ZRB06182, is a recombinant Rabbit Monoclonal Antibody that detects p44 MAPK (ERK1) and has been tested for use in Immunohistochemistry (Paraffin), Immunoprecipitation, and Western Blotting.
Western Blotting Analysis: 1:1,000 dilution from a representative lot detected p44 MAPK (ERK1) in Rat C6, L6, and mouse NIH/3T3 cell lysates.

Immunoprecipitation Analysis: 100 μL from a representative lot immunoprecipitated p44 MAPK (ERK1) in Rat L6 cell lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected p44 MAPK (ERK1) in rat colon and rat cerebral cortex tissue sections.

Target description

Mitogen-activated protein kinase 3 (UniProt: P21708; also known as EC: 2.7.11.24, MAP kinase 3, MAPK 3, ERT2, Extracellular signal-regulated kinase 1, ERK-1, Insulin-stimulated MAP2 kinase, MAP kinase isoform p44, p44-MAPK, MNK1, Microtubule-associated protein 2 kinase, p44-ERK1) is encoded by the Mapk3 (also known as Erk1, Prkm3) gene (Gene ID: 50689) in rat. ERK-1 is a serine/threonine kinase that acts as an essential component of the MAP kinase signal transduction pathway. It is expressed in different tissues, mostly in intestine, placenta, and lung. Highest levels have been reported in the nervous system. The MAPK/ERK cascade mediates several diverse biological processes such as cell growth, adhesion, survival, and differentiation through the regulation of transcription, translation, and cytoskeletal rearrangements. It also plays also a role in initiation and regulation of meiosis, mitosis, and post-mitotic functions in differentiated cells by phosphorylating a few transcription factors. About 160 substrates have been described for ERKs. The protein kinase domain of ERK-1 is localized to amino acids 43-331 and the TXY motif is located in amino acids 203-205. ERK-1 is phosphorylated by MEK1 and MEK2 on threonine 203 and tyrosine 205 in response to external stimuli like insulin or nerve growth factor and both these phosphorylations are essential for its activity. Two isoforms of ERK-1 have been described that are produced by alternative splicing. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
