General description

NDRG3 (N-myc downstream-regulated gene 3 protein) is a novel member of N-myc down-regulated gene (NDRG) family mapped to human chromosomal location 20q11.21-q11.23. It is abundantly expressed in brain, prostate, ovary, and testis tissues. It is localized to the outer layers of seminiferous epithelium.

Biochem/physiol Actions

N-myc downstream-regulated gene 3 protein (NDRG3) is involved in prostate cancer cell growth in vitro and in vivo. It acts as an androgen regulated and prostate enriched gene during controlling of cancer cell growth. Thus, overexpression of NDRG3 has been linked to the malignant phenotype of tumors. In hepatitis B virus (HBV)-related hepatocellular carcinoma cell line, NDRG3 gene is inversely expressed with the miR-122 expression. Thus, it has been reported that in conjugation with miR-122, NDRG3 can be a potential therapeutic marker for hepatocellular carcinoma (HCC). Prominent localization of NDRG3 in the epithelium cells indicate its role in spermatogenesis. It also plays an important role in lactate-induced hypoxia signaling pathway in hypoxia. Its protein level expression in cell is negatively regulated by oxygen, whereas, whereas excess lactate produced under prolonged hypoxia blocks its proteasomal degradation by binding to NDRG3.
