General description

The GlycoProfile IV Kit utilizes trifluoromethane sulfonic acid (TFMS) in a deglycosylation system that completely removes all N- and O-linked glycans while safeguarding the protein or polypeptide structure. TFMS hydrolysis of glycoproteins results in minimal protein degradation while the released glycans are eliminated. The conditions are optimized to deglycosylate simple or low molecular weight glycoproteins in 30 mins without the scavenger. For high molecular weight or complex, non-mammalian glycoproteins, anhydrous anisole is provided as a scavenger to give a high protein yield.

Application

GlycoProfile IV Chemical Deglycosylation Kit has been used in the deglycosylation of human α-L-iduronidase and peritrophic matrix from the gut of the caterpillar, Helicoverpa armigera. It has also been used to estimate the glycosylation of plant-derived recombinant human interleukin-13 (rhIL-13).

Quantity

The kit contains sufficient reagents for 10 reactions with a sample size between 1-2 mg of a typical glycoprotein. An RNase B glycoprotein standard is included.
