General description

Transcription factor E2F1 (UniProt: Q01094; also known as E2F-1, PBR3, Retinoblastoma-associated protein 1, RBAP-1, Retinoblastoma-binding protein 3, RBBP-3, pRB-binding protein E2F-1) is encoded by the E2F1 (also known as RBBP3) gene (Gene ID: 1869) in human. E2F-1 is a transcription activator that binds DNA cooperatively with DP proteins through the E2 recognition site, 5′-TTTC[CG]CGC-3′ found in the promoter region of a number of genes whose products are involved in cell cycle regulation or in DNA replication. It is a component of the DRTF1/E2F transcription factor complex and forms heterodimers with DP family members. The DRTF1/E2F complex functions in the control of cell-cycle progression from G1 to S phase. The E2F1 complex is shown to bind specifically hypophosphorylated retinoblastoma protein RB1. During the cell cycle, RB1 becomes phosphorylated in mid-to-late G1 phase and detaches from the DRTF1/E2F complex that renders E2F transcriptionally active. E2F-1 can be phosphorylated by CDK2 and cyclin A-CDK2 in the S-phase. It is also phosphorylated at serine 364 by CHEK2, which stabilizes it upon DNA damage and regulates its effect on transcription and apoptosis. Acetylation of E2F-1 by P/CAF/KAT2B stimulates its DNA-binding.

Specificity

Clone 71-10 is a rabbit monoclonal antibody that specifically detects human E2F1 phosphorylated on Serine 375.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids surrounding phosphoserine 375 from the C-terminal region of human transcription factor E2F1.

Application

Anti-phospho-E2F1 (Ser375), clone 71-10, Cat. No. MABE1782, is a highly specific rabbit monoclonal antibody that targets Transcription factor E2F1 phosphorylated on Serine 375 and has been tested in ELISA and Western Blotting.
ELISA Analysis: A representative lot detected phospho-E2F1 (Ser375) in ELISA applications (Dale, T., et. al. (2015). Nat Chem Biol. 11(12):973-980).

Quality

Evaluated by Western Blotting in HEK293 cells transfected with pcDNA3.1 empty vector, HEK 293 cells transfected with E2F1 wild type-GST, HEK 293 cells transfected with E2F1 wild type-GST with alanine substituted for serine at 375.

Western Blotting Analysis: A 1:2,000 dilution of this antibody detected phospho-E2F1 (Ser375) in HEK293 cells transfected with E2F1 wild type-GST,, but not in HEK 293 cells transfected with E2F1 wild type-GST with alanine substituted for serine at 375.

Target description

~80 kDa observed ( GST fusion protein); 46.92 kDa calculated (without GST). Uncharacterized bands may be observed in some lysate(s).
