General description

Glucocorticoid receptor (GR), also known as Nuclear receptor subfamily 3 group C member 1, and encoded by the gene name NR3C1/ GRL, is receptor for glucocorticoids (GC). GR has a dual mode of action: as a transcription factor that binds to glucocorticoid response elements (GRE), both for nuclear and mitochondrial DNA, and as a modulator of other transcription factors. GR affects inflammatory responses, cellular proliferation and differentiation in target tissues. Recent studies have shown that GR may act as a coactivator for STAT5-dependent transcription upon growth hormone (GH) stimulation and could reveal an essential role of hepatic GR in the control of body growth. In addition, GR is involved in chromatin remodeling, and may play a negative role in adipogenesis through the regulation of lipolytic and antilipogenic genes expression. Mutation in GR has been associated with Glucocorticoid resistance (GCRES): a hypertensive, hyperandrogenic disorder characterized by increased serum cortisol concentrations. Inheritance is autosomal dominant. GR is widely expressed. In the heart, GR is detected in left and right atria, left and right ventricles, aorta, apex, intraventricular septum, and atrioventricular node, as well as whole adult and fetal heart.
