General description

Direct Red 80 (C.I. 35780) is an extremely hydrophilic, very large tetrakisazo hexasulfonated anionic dye that has a large, planar aromatic system.It is also known as Sirius red F3B, chlorantine fast red 7BLN or 5BRL, durazol brilliant red B, pontamine fast red 7BNL, and sirius fast red.

Application

Direct Red 80 has been used as an ingredient in the preparation of saturated picric acid for immunohistochemical detection and quantitation of collagen in liver tissues.Other applications include -
  • detection of amyloid
  • Sirius red-Van Gieson methods for quantitation of collagen using image analysis methods
  • counterstaining applications substituting eosin in alum hematoxylins

  1. Is Direct Red 80 comparable to Dextran Red for back-filling injections in live zebrafish spinal cord?

    1. Direct Red 80 is more commonly referred to as Sirius Red and is a stain for collagen. It is a vital stain used for various types of biological staining. Dextran Red is a fluorescent dye for which you would require a fluorescent microscope to view the staining. The different molecular weights of dextran are used to determine whether the dye crosses certain barriers.

  2. Using Direct Red 80 with Picric acid P6744 for Picro Sirius Red staining, and the staining resultss are coming out very yellow. Any advice on this issue?

    1. Direct Red 80 is not performance tested for use in a Picro Sirius Red staining method. Tissues fixed or stained with Picric Acid may require long rinse or wash steps to remove excess picric acid, and using slightly acidified water has been seen in protocols to remove the excess picric acid from tissue sections.
