General description

1,2 ARTS was later shown to be a key apoptotic protein mediating apoptosis initiated by other inducers.
Anti-ARTS antibody, Mouse monoclonal (mouse IgG2b isotype) is derived from the ARTS51 hybridoma produced by the fusion of mouse myeloma cells and splenocytes from a BALB/c mouse immunized with a synthetic peptide corresponding to C-terminus of human ARTS protein, conjugated to KLH. ARTS (Apoptosis Related protein in the TGF-β signaling pathway), is a novel protein involved in the apoptotic pathways of transforming growth factor-β (TGF-β). The protein is derived from the human septin H5/PNUTL2/CDCrel-2a/2b gene. ARTS consists a phosphate-binding site (P-loop) death domain, which is conserved in all septins and in many different classes of ATP/GTPases. It is localized to mitochondria and translocates to the nucleus, coincident with apoptotic signals induced by TGF-β.
Apoptosis-related protein in TGF-β signaling pathway (ARTS) is a novel mitochondrial protein that mediates apoptosis induced by a variety of pro-apoptotic stimuli. Originally found to be essential for apoptosis induced by transforming growth factor-β (TGF-β) ARTS was later shown to be a key apoptotic protein mediating apoptosis initiated by other inducers. Mouse monoclonal anti-ARTS antibody recognizes an epitope within amino acids 247-266 of human ARTS molecule.

Specificity

This monoclonal anti-ARTS antibody was used to detect ARTS (Sept4_i2) protein in whole lysates of HeLa cells. HeLa ARTS knockdown (ARTS KD) stable cell line was established by using short hairpin RNAs (shRNAs) which abrogated specifically the endogenous levels of ARTS (Sept4_i2, ~30Kd)

This monoclonal Ab is the only currently commercially available antibody directed against the unique C-terminus of ARTS, that can distinguish betweenARTS ( the pro-apoptotic, tumor suppressor, specific XIAP- antagonist) and Sept4_i1 (H5,PNUTL), which is the other product of the SEPT4 gene.

Immunogen

synthetic peptide corresponding to C-terminal region (amino acids 247-266) of human ARTS protein, conjugated to KLH.

Application

Immunoprecipitation of COS-7 cell lysates was performed using 10 μl of monoclonal mouse anti-ARTS antibody. Antibody was allowed to bind for 4 hours on ice before precipitated using G-sepharose beads.
Monoclonal anti-ARTS antibody has been used for western blot assays. The antibody has also been used for immunostaining applications. Furthermore, the product is suitable for use in indirect ELISA microarray and immunocytochemistry assays.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
