General description

T-cell surface glycoprotein CD3 zeta chain (CD3ζ), also called CD247, is a component of the T-cell antigen receptor (TCR) multimeric complex. CD247 gene is mapped to human chromosome1q24.2 and is majorly expressed in the natural killer and T cells. CD3ζ comprises three immunoreceptor tyrosine-based activation motifs (ITAMs), the short ectodomain of nine amino acids and a long cytoplasmic tail region. The ITAMs possess conserved Tyr-X-X-Leu/lle sequence.

Specificity

Anti-phospho-CD3z(pTyr83) recognizes mouse phosphorylated CD3z(pTyr83).

Immunogen

synthetic phosphopeptide corresponding to residues surrounding Tyr83 of mouse CD3ζ, conjugated to KLH. The corresponding sequence differs by a single amino acid in human.

Application

Anti-phospho-CD3ζ (pTyr83) antibody produced in rabbit may be used in immunoblotting and immunoprecipitation.

Biochem/physiol Actions

T-cell surface glycoprotein CD3 zeta chain (CD3ζ or CD3z) is involved in the activation of T cells.. The signal peptide in the N-terminal region aids in targeting protein translation to the endoplasmic reticulum. It plays a key role in the recognition of the antigenic peptide that is presented by the human leukocyte antigen on antigen-presenting cells or tumor cells. CD3ζ chain plays a key role in regulating TCR assembly, transport, and cell surface expression. It reorganizes the actin-based cytoskeleton and regulates the morphology of immune cells. The absence of CD3ζ in T cells is postulated to be involved in the pathology of rheumatoid arthritis, systemic lupus erythematosus (SLE) and cancer. Lack of CD3ζ impairs retinal ganglion cell (RGC) motility and its presence is crucial for synaptic wiring in the retina. Downregulation of CD247 also impacts the expression of the zeta chain of T cell receptor associated protein kinase 70 (ZAP70) of the T-cell signal-transduction pathway. It is regarded as a potential therapeutic target for ovarian cancer treatment.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
