General description

IgG (Immunoglobulin G) antibody subtype is the most abundant of serum immunoglobulins of the immune system. It is secreted by B cells and is found in blood and extracellular fluids. Maternal IgG is transferred to fetus through the placenta that is vital for immune defense of the neonate against infections.
Immunoglobulins have two heavy chains and two light chains connected by disulfide bond. It is a glycoprotein which mainly helps in immune defense. IgG is a major class of immunoglobulin. Rat has five immunoglobulin classes- IgM, IgG, IgA, IgD and IgGE. IgG has four subclasses- IgG1, IgG2a, IgG2b and IgG2c.

Specificity

Anti-Rat IgG (whole molecule) antibody recognizes rat (immunoglobulin G) IgG and is determined by immunoelectrophoresis (IEP) versus normal rat serum and rat IgG prior to conjugation.

Application

Anti-Rat IgG (whole molecule) Peroxidase antibody produced in rabbit has been used in western blotting and enzyme linked immunosorbent assay (ELISA).

Biochem/physiol Actions

IgGs are glycoprotein antibodies that modulate several immune responses. Rat IgGs against target proteins are often used as primary antibodies in various research applications. Thus, secondary anti-rat IgGs conjugated to a detectable substrate are useful tools for the analysis of target proteins.
Immunoglobulin G (IgG) participates in hypersensitivity type II and type III. It helps in opsonization, complement fixation and antibody dependent cell mediated cytotoxicity.

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 0.05% MIT

Preparation Note

Prepared by the two-step glutaraldehyde method described by Avrameas, S., et al., Scand. J. Immunol., 8, Suppl. 7, 7 (1978).

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage, the solution may be frozen in working aliquots. Repeated freezing and thawing, or storage in "frost-free" freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use.
