Specificity

Clone 1J23 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects RNA-binding protein MEX3A. It targets an epitope within 16 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 16 amino acids from the C-terminal half of human RNA-binding protein MEX3A.

Application

Quality Control Testing

Evaluated by Western Blotting in SH-SY5Y cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected MEX3A in SH-SY5Y cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected MEX3A in Neuro2A and U87MG cell lysates.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected MEX3A in SH-SY5Y cells.

Affinity Binding Assay: A representative lot of this antibody bound MEX3A peptide with a KD of 1.0 x 10-12 in an affinity binding assay.

Target description

RNA-binding protein MEX3A (UniProt: A1L020; also known as RING finger and KH domain-containing protein 4) is encoded by the MEX3A (also known as RKHD4) gene (Gene ID: 92312) in human. MEX3A is a member of the MEX3 family that widely exists in mammalian cells and encodes four different genes, MEX3A, MEX3B, MEX3C, and MEX3D. These four genes are located on distinct chromosomes and encode closely related proteins. MEX3A is an RNA-binding phosphoprotein that is involved in post-transcriptional regulatory mechanisms. It is predominantly expressed in the cytoplasm and shuttles between the cytoplasm and the nucleus through the CRM1 export pathway. Its highest levels are observed in fetal brain and testis. Its expression is also observed in thymus, salivary gland, and uterus. MEX3A contains two RNA-binding KH domains (aa 132-193 and223-284) and a ring domain with E3 ubiquitin ligase activity at the C terminal. The ring finger domain provides MEX3 protein the ability to regulate ubiquitination of target proteins and influencing their protein stability and subcellular localization. Overexpression of MEX3A is reported in gliomas and is linked to higher malignancy and poor prognosis. In vitro loss-of-function and gain-of-function studies show that MEX3A promotes glioma development through regulating cell proliferation, cell apoptosis, cell cycle, and cell migration. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
