Specificity

Clone 1B11 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects GATA-3. It targets an epitope within 21 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the N-terminal half of human GATA-3.

Application

Quality Control Testing

Evaluated by Western Blotting in SH-SY5Y cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected GATA-3 in SH-SY5Y cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected GATA-3 in Jurkat and Neuro2A cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected GATA-3 in human breast cancer tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound GATA-3 with a KD of 1.5 x 10-7 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:300 dilution from a representative lot detected GATA-3 in MCF-7 cells.

Target description

Trans-acting T-cell-specific transcription factor GATA-3 (UniProt: P23771; also known as GATA-binding factor 3) is encoded by the GATA3 gene (Gene ID: 2625) in human. The GATA family of transcription factors contain one or two C2 C2-type zinc-finger motifs that recognize the consensus DNA sequence WGATAR (W denotes A or T, and R denotes A or G). Six members of this family have been described in mammals. GATA1, 2, and 3 are hematopoietic factors and GATA4, 5, and 6 are considered as endodermal factors. They display a common structure, which comprises distinct amino-terminal regions with transactivating activity, highly conserved zinc-finger motifs in the carboxy-terminal region, conserved basic regions that are located immediately after the second zinc finger, and distinct C-terminal regions of varying lengths. GATA-3 is a highly conserved member of the GATA family with two zinc-finger motifs (aa 263-287 and 317-341). It is expressed in T cells and plays a key role in the control of CD4+ effect T cell differentiation. Its expression is also observed in endothelium, kidney, fetal liver, and nervous tissues. It is continually expressed in a highly regulated manner throughout T-cell development and CD4+ T helper (TH)-cell differentiation. It is shown to be crucial for commitment to the T lineage at the point when hematopoietic common lymphoid progenitors (CLPs) destined to become B cells diverge from those destined to become T cells. GATA3 mutations in breast tissue are associated with several tumor characteristics and influence the pattern of gene expression and these mutations appear to be of prognostic value in estrogen receptor (ER)-positive subjects. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
