General description

Anti-Glycogen Synthase Kinase-3β (GSK-3β) is developed in rabbit using a synthetic peptide (KGDRGQTNNAASASASNST) corresponding to the C-terminal region of human GSK-3β conjugated to KLH as immunogen. Glycogen Synthase Kinase-3β (GSK-3β) is present in cytoplasm, nucleus and mitochondria. GSK-3 exists as two closely related protein kinases, termed GSK-3α and GSK-3β. This protein contains a protein kinase domain in its core, and is composed of 482 amino acids. It shows a wide range of tissue expression with the highest expression in brain.
Glycogen synthase kinase-3 β (GSK3β) is a 47kD protein that belongs to serine/ threonine protein kinase family and is expressed ubiquitously as a constitutively active kinase. It has a pivotal role in regulating the degradation of β-catenin by the tumor suppressor gene product APC. Anti-Glycogen Synthase Kinase-3β (GSK-3β) antibody can be used in microarray. Rabbit anti- GSK3β antibody reacts specifically with rat GSK3β but not with GSK3α.

Immunogen

Synthetic peptide corresponding to amino acids 403-420 of human glycogen synthase kinase 3 β (GSK3β), coupled to KLH. It is identical in rat and highly conserved in frog and zebrafish GSK3β.

Application

Anti-Glycogen Synthase Kinase-3β (GSK-3β) antibody produced in rabbit has been used in immunoblotting assays for analysis of protein expression.

Biochem/physiol Actions

Glycogen synthase kinase-3 (GSK-3) specifically phosphorylates glycogen synthase, a critical enzyme regulating glucose storage, and mediates insulin regulation of glycogen synthesis.(8) GSK-3β phosphorylates regulatory sites in the central region of APC. Binding of β-catenin to APC, marking β-catenin for destruction, is dependent on phosphorylation of APC by GSK-3β. Thus, GSK-3β may play a critical role in regulating β-catenin stability during development and tumor progression. It is responsible for rapid cyclin D1 turnover by phosphorylating it on Thr-286. It functions as a negative regulator of Wnt signaling during embryonic development and cell proliferation in adult tissues.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 1% bovine serum albumin and 15 mM sodium azide.
