General description

PSD-95 is part of a multiprotein complex restricted to the post-synaptic membrane. It is believed to initiate NMDA receptor clustering on the post synaptic membrane. Its restricted expression makes it an excellent marker for post-synaptic membrane regions on neurons.

Specificity

Other species have not been tested.
Reacts with a protein of ~100 kDa, corresponding to the apparent molecular weight of PSD-95, on SDS-PAGE immunoblots of samples from rat, mouse and bovine. A slightly higher molecular weight band is also detected. In rat brain and mouse brain tissue extracts a band of ~75 kDa is also present.

Confocal fluorescent analysis of PC12 using MAb1596 Mouse mAB (Red). Actin filaments have been labeled with Alexa Fluor 488 -Phalloidin (Green). Nuclear is stained with DAPI (Blue). Membrane staining positive.

Immunocytochemistry:
12.5 µg/mL

Western Blot:
1-5 μg/mL (ECL) on rat brain lysate (20 μg).

Optimal working dilutions must be determined by the end user.

Storage and Stability

Stable for 6 months at -20°C in undiluted aliquots from date of receipt. Do not store below -20°C.
Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variabillity in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.
