General description

Monogalactosyldiacylglycerol (MGDG) and digalactosyldiacylglycerol (DGDG) are the two nonionic lipid constituents of the thylakoid membrane of higher plant chloroplasts. MGDG and DGDG are present in the membrane at 56% and 29%, respectively, of the total lipid content. DGDG is a bilayer-forming lipid, while MGDG alone will only form hexagonal-II structures.
Monogalactosyldiacylglycerol (MGDG) synthesis involves generation of monoglucosyldiacylglycerol (MGlcDG) from diacylglycerol (DAG) in the presence of MGlcDG synthase. The second step is epimerization of MGlcDG to MGDG. It forms nonbilayer structures and is also associated with algae and fungi.

Application

Monogalactosyldiacylglycerol (MGDG) may be used:
  • in bicelles preparation for nuclear magnetic resonance studies
  • in the generation of structured MGDG for pinolenic acid enrichment
  • in the preparation of liposomes
  • as a lipid standard in hydrophilic interaction liquid chromatography-ion mobility mass spectrometry (HILIC-IM-MS)

Biochem/physiol Actions

Monogalactosyldiacylglycerol (MGDG) synthesis is essential for chloroplast biogenesis. It is also needed for synchronizing photosynthesis associated plastid and nucleus related gene expression. MGDG is essential for membrane lipid homeostasis during stress. In food industry, MGDG serves as an enhancer in baking and as an non-ionic emulsifier.

Galactolipids are the main structural component of plant chloroplastic (thylakoid) membranes and of blue-green algae cell membranes. The predominant lipids in this class are monogalactosyl-diacylglycerol (MGDG) and digalactosyl-diacylglycerol (DGDG). We here present a method for the preparation of bicelles that
