Specificity

Clone 4L14 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Methylcytosine dioxygenase TET2. It targets an epitope within the N-terminal region.

Immunogen

His-tagged recombinant fragment corresponding to the first 300 amino acids from the N-terminal region of human Methylcytosine dioxygenase TET2.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected TET2 in HeLa cell lysate.

Tested applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected TET2 in TF-1, L6, and NIH3T3 cell lysates.

Affinity Binding Assay: A representative lot of this antibody bound recombinant TET2 fragment with a KD of 4.1 x 10-7 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TET2 in HeLa, HUVEC, and NIH3T3 cells.

Anti-TET2, clone 4L14 ZooMAb®, Cat. No. ZRB1395, is a recombinant Rabbit monoclonal antibody that detects TET2 and is tested for use in Affinity Binding Assay, Immunocytochemistry, and Western Blotting.

Target description

Methylcytosine dioxygenase TET2 (UniProt: Q6N021; also known as EC:1.14.11.n2, TET2) is encoded by the TET2 (also known as KIAA1546, Nbla00191) gene (Gene ID: 54790) in human. TET2 is an evolutionarily conserved nuclear protein with several Fe2+ and Zn2+ binding sites. It is highly expressed in hematopoietic stem/progenitor cells and serves as a dioxygenase that catalyzes the conversion of the modified genomic base 5-methylcytosine (5mC) into 5-hydroxymethylcytosine (5hmC) and plays a key role in active DNA demethylation. It displays a preference for 5-hydroxymethylcytosine in CpG motifs. TET2 also mediates subsequent conversion of 5hmC into 5-formylcytosine (5fC), and conversion of 5fC to 5-carboxylcytosine (5caC). Methylation at the C5 position of cytosine bases is an epigenetic modification of the mammalian genome which plays an important role in transcriptional regulation. In addition to its role in DNA demethylation, it is also involved in the recruitment of the O-GlcNAc transferase OGT to CpG-rich transcription start sites of active genes that helps in promoting histone H2B GlcNAcylation by OGT. TET2 is reported to be frequently mutated in myeloproliferative disorders that involve cellular proliferation of one or more hematologic cell lines in the peripheral blood, distinct from acute leukemia. These disorders include essential thrombocythemia, polycythemia vera, and primary myelofibrosis. Deletion of TET2 in primary bone marrow cells is shown to increase the percentage of immature c-Kit+ Lin cells, indicating that loss of TET2 can affect stem/progenitor cell differentiation. Three isoforms of TET2 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
