General description

Ubiquitin-specific peptidase 18 (USP18), an isopeptidase codes for a protein with a molecular mass of 43 KDa, UBP43. The promoter region of USP18 gene possesses interferon-sensitive response elements (ISREs). USP18 protein has cysteine and histidine boxes, that are responsible for enzymatic activity. The expression of USP18 is seen in the liver, thymus, and spleen. It is also expressed at a low level in adipose tissue, lung, and bone marrow. The USP18 gene is located on human chromosome 22q11.2.

Biochem/physiol Actions

Ubiquitin-specific peptidase 18 (USP18) serves as a modulator of interferon type 1 (IFN-I) signaling. It plays a major role in stress responses and signal transduction pathways. This protein is also involved in responses to the infection of virus and bacteria. Through interferon stimulated gene 15 (ISG15) (ISGylation), it can reverse the post-translational modifications (PTM) of target proteins.
