Cell Line Description
Flat ribbon-like cells differentiating to parallel arrays of spindle shaped cells. Derived from the thoracic aorta of an embryonic BDIX rat. As cells achieve the stationary phase there is an increase in myokinase and creatine phosphokinase (CPK) activity. After cessation of cell division a muscle type CPK isoenzyme is synthesised.
Application
A7r5 Cell Line from rat has been used to study the effect of angiotensin II on vascular oxidative stress. It has been used in PLA2 inhibitory study to understand the role of PLA2 on formation of lipid droplets.
Biochemical and biophysical studies
Culture Medium
DMEM + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).
Subculture Routine
Split sub-confluent cultures (70-80%) 1:2 to 1:3 i.e. seeding at 3-5x10,000 cells/cm2 using 0.25% trypsin; 5% CO2; 37°C. After resuscitation cells may take up to 7 days before a monolayer forms and growth can be very slow in general. Cells also grow in clumps.
