General description

ASF1 is a histone chaperone protein that binds H3 and H4. It functions in nucleosome assembly.
Monoclonal Anti-ASF1 (mouse IgG2a isotype) is derived from the hybridoma 4A1/3 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with an ASF1A fusion protein. ASF1 (anti-silencing function 1) encodes a member of the H3/H4 family of histone chaperone proteins and is similar to the anti-silencing function-1 gene in yeast.

Monoclonal Anti-ASF1 antibody produced in mouse has been used in immunoblotting.

Biochem/physiol Actions

ASF1 (anti-silencing function 1) protein is a key component of a histone donor complex. It interacts with histones H3 and H4 and functions together with a chromatin assembly factor 1 (CAF-1) during DNA replication and repair. ASF1 also contributes to genome stability during S phase. Cells lacking ASF1 were found to be sensitive to a number of genotoxic agents that predominantly cause DNA damage by interfering with DNA replication fork progression such as hydroxyurea and camptothecin.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
