Brain derived neurotrophic factor (BDNF) belongs to a family of secreted proteins called neurotrophins that includes, nerve growth factor (NGF), neurotrophin-3 (NT3) and NT4/5. BDNF is expressed in the developing and the mature brain. The influence of BDNF is mediated by its binding to TrkB receptor and sometimes the receptor p75NTR. The downstream pathways promoted by BDNF include PI3K, MAPK and JAK/STAT. The most important role of BDNF is the regulation of synaptic transmission and plasticity, protein synthesis and phosphorylation of local proteins. The secretion of BDNF is highly regulated at both transcriptional and translational levels since it is involved in the proper function of neuronal circuit and development of cognitive functions.
