Gene Information

human ... PCM1(5108)

General description

Pericentriolar material 1 (PCM1) is a centriolar satellite protein. The PCM1 gene is located on the human chromosome at 8p22.

Biochem/physiol Actions

Pericentriolar material 1 (PCM1) protein acts as a scaffold and facilitates the aggregation of target proteins to the centrosome during centrosome maturation. It forms a complex with Bardet-Biedl syndrome 4 protein (BBS4) and disrupted-in-schizophrenia 1 (DISC1) at the centrosome. PCM1 along with DISC1 and BBS4 plays a role in the centrosomal pathway that is associated with schizophrenia. It is involved in ciliogenesis by binding an E3 ligase enzyme to satellites. PCM1 regulates the primary cilia disassembly before mitotic entry by recruiting polo-like-kinase 1 (Plk1) to the pericentriolar matrix.

Primary cilia, which emanate from the cell surface, exhibit assembly and disassembly dynamics along the progression of the cell cycle. However, the mechanism that links ciliary dynamics and cell cycle regulation remains elusive. In the present study, we report that

To elucidate the role of centriolar satellites in ciliogenesis, we deleted the gene encoding the PCM1 protein, an integral component of satellites. PCM1 null human cells show marked defects in ciliogenesis, precipitated by the loss of specific proteins from satellites
