Application

Alkaline phosphatase is used in the laboratory to remove phosphate monoesters which prevents self-ligation. It is also used in the dairy industry to confirm pasturization of milk. Alkaline phosphatse is commonly conjugated to antibodies and used in immunoblotting, ELISA′s, and other antibody based detection assays.

Biochem/physiol Actions

Alkaline phosphatase is a hydrolytic enzyme which dephosphorylates a variety of molecules under basic conditions.

Unit Definition

1 U corresponds to the amount of enzyme which hydrolyzes 1 μmol 4-nitrophenyl phosphate per minute at pH 9.8 and 37 °C

Reconstitution

The solution can be used without dialysis (for enzyme immuno-assay).

Immunoliposomes were prepared and their immunoassay applications investigated. Liposomes were prepared from cholesterol and phospholipids including maleimidobenzoylphosphatidylethanolamine (MBPE) for conjunction with thiol-containing antigens. Alkaline phosphatase (APase) was entrapped in the liposome and BSA, the antigen, was modified by reaction with
