Leptin (Ob) is a non-glycosylated polypeptide hormone. It is produced primarily in adipocytes and the placenta. The structure of leptin closely resembles members of the long-chain helical cytokine family. It is composed of four antiparallel α-helices interconnected by two long crossover links and one short loop. These structural elements are arranged in a left-handed helical bundle formation, resulting in a unique two-layer packing configuration.

Application

Leptin from mouse has been used:
  • to check the effect of leptin on ovulation for treating infertility in obese mice
  • to check the effect of leptin on MGARP (mitochondria-localized glutamic acid-rich protein) expression in testis and follicular granular cells of mice
  • to study the morphometrical changes in mouse testicular tissue upon long term exposure to leptin

Biochem/physiol Actions

Leptin is a hormone produced primarily in adipocytes, although leptin mRNA has also been identified in placenta and fetal tissues, gastric tissue and liver. Its primary site of action appears to be on neurons in the hypothalamus that are involved in regulating energy balance, appetite, and body weight. Leptin increases the production of nitric oxide in endothelial cells and stimulates angiogenesis in vitro and in vivo.
Human and mouse leptin share ~84% sequence identity.
Leptin, an essential cytokine, serves as a signaling molecule in the reproductive system. It modulates placental functions through autocrine or paracrine signaling. Particularly in the early stages of pregnancy, leptin plays a vital role in modulating key processes including cell proliferation, protein synthesis, invasion, and apoptosis within placental cells. Deficiency of leptin may lead to the development of severe obesity and abnormalities in hematopoiesis, immunity, angiogenesis, bone formation, blood pressure regulation and reproduction. Absence of leptin in mice causes severe obesity and hypothermia at temperature below thermoneutrality. It also controls feeding behavior. Leptin signaling is crucial for the bone formation.

The biological activity is measured by its ability to induce proliferation of leptin-dependent human OB-R transfected murine BAF3 cells.
