General description

APEX1 (apurinic/apyrimidinic endodeoxyribonuclease 1) is a multifunctional DNA base excision repair (BER) protein, associated with various cellular functions including oxidative signaling, transcription factor regulation and cell cycle control.

Application

The Human Protein Atlas project can be subdivided into three efforts: Human Tissue Atlas, Cancer Atlas, and Human Cell Atlas. The antibodies that have been generated in support of the Tissue and Cancer Atlas projects have been tested by immunohistochemistry against hundreds of normal and disease tissues and through the recent efforts of the Human Cell Atlas project, many have been characterized by immunofluorescence to map the human proteome not only at the tissue level but now at the subcellular level. Biochem/physiol Actions

APEX1 (apurinic/apyrimidinic endodeoxyribonuclease 1) performs an essential role in the DNA BER (base excision repair) pathway. It acts on the damaged bases caused by endogenous and exogenous agents. It accelerates the cleaving of 5′ end, an abasic site in duplex DNA, through Mg2+-dependent manner. It stimulates the DNA-binding activity of transcription factors such as Fos-Jun, nuclear factor-κB, Myb, HLF, PAX, and p53. It is also involved in the apoptosis associated with thioredoxin. It is upregulated in various cancer cells, such as germ cell tumors (GCTs) in testicular cancer. It acts as a ′hub-protein′ that controls pathways essential for the cell survival, such as energy metabolism, cytoskeletal elements and stress-dependent responses.
