General description

Synapsin I is a member of the synapsin family. Synapsins are neuronal phosphoproteins which associate with the cytoplasmic surface of synaptic vesicles. Family members are characterized by common protein domains, and they are implicated in synaptogenesis and the modulation of neurotransmitter release, suggesting a potential role in several neuropsychiatric diseases. This member of the synapsin family plays a role in regulation of axonogenesis and synaptogenesis. The protein serves as a substrate for several different protein kinases and phosphorylation may function in the regulation of this protein in the nerve terminal. Mutations of the Synapsin I gene may be associated with X linked disorders with primary neuronal degeneration such as Rett syndrome.

Specificity

Synapsin I (Mr of 80,000 and 77,000). Immunolabeling is blocked by preadsorption of antibody with Synapsin I. Synapsin is located only in brain nerve terminal and is an excellent marker for synapses.

Immunogen

Synapsin I (mixture of Ia & Ib) purified from bovine brain.

Western blotting:
1:200 of a previous lot was used for labeled Protein A, 1:1,000 dilution for ECL.

Immunocytochemistry:
1:500-1:2,000 dilution of a previous lot was used.

Immunoprecipitation:
5 μL of a previous lot quantitatively immunoprecipitated all synapsin in 200 μg of rat brain.

Optimal working dilutions must be determined by the end user.

Quality

Immunohistochemistry(paraffin): Synapsin representative staining pattern/morphology in rat hippocampal neurons. Tissue was pretreated with Citrate pH 6.0, antigen retrieval. A previous lot of this antibody was diluted to 1:1000, using IHC-Select reagents used with HRP-DAB. Immunoreactivity is seen directly associated with terminal end of neuronal cell body.

Stable for 1 year at -20ºC from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
